Highlights from this day
  • blind_submit — genuine question changed my mind on this, i started thinking amylin was the interesting bit and now i think its the pairing, ill dig out the number 09:20
  • blind_submit — the combination is the thing with trial data behind it, monotherapy in here is mostly us guessing 10:57
  • tallinn_taper — dropped the GLP-1 and stayed on this alone for 10 months, appetite held but weight barely moved 11:18
  • tallinn_taper — noticed something around week 32 on monotherapy, well after i had given up expecting to 12:36
  • tallinn_taper — added it to an existing tirzepatide dose rather than starting both, which made attribution possible, happy to be corrected 12:39

stopped for 16 months and the fullness went within a fortnight of the last dose

🧪8⚠️1😂9

combination GI was worse than either alone for me, which is not what i had read anywhere

TT

did anyone add this to an existing tirzepatide dose rather than starting both

dropped the GLP-1 and stayed on this alone for 10 months, appetite held but weight barely moved

if youre used to GLP-1 titration steps this feels absurdly cautious and i still do it that way

TT

noticed something around week 32 on monotherapy, well after i had given up expecting to

its an amylin analogue, different receptor family, so treating it as another GLP-1 is the first mistake

added it to an existing tirzepatide dose rather than starting both, which made attribution possible, happy to be corrected

held mine a while

Cited study
Efficacy and Safety of Cagrilintide in Adults with Overweight or Obesity — Phase 2
The Lancet · 2021
Cagrilintide monotherapy, dose ranging. The combination trials came later.

REDEFINE is the phase 3 programme, most of what gets quoted casually in here is earlier work, i could be wrong

amylin is about slowed gastric emptying and satiety signalling, thats the short version and im no pharmacologist, ymmv

AH

i titrated over twice as many weeks as i did on semaglutide and that was right for me
