is there any point splitting an oral dose

- thirty_min_wait — orforglipron is a small molecule rather than a peptide, which is why the fasting conditions differ, happy to be corrected 20:12
- thirty_min_wait — back after 26 months, has anything changed on the oral side i use it because needles are a genuine problem for me, not because i think its better 20:30
- batch_bandit — ok so PIONEER was the original oral semaglutide programme and OASIS looked at the higher oral doses lost 25kg over 1 months on oral, slower than my injectable… 21:03
- typosquat_tay — roughly one percent gets absorbed, which is why the tablet strength looks nothing like an injectable dose, thats just me 21:41
orforglipron is a small molecule rather than a peptide, which is why the fasting conditions differ, happy to be corrected
is the absorption variability why the oral doses look so high
not a peptide
i use it because needles are a genuine problem for me, not because i think its better
nobody should copy my morning routine, but the timer is the bit i would keep
absorption is the variable, not the molecule. same tablet, different morning, different result
[edited]back after 26 months, has anything changed on the oral side
i use it because needles are a genuine problem for me, not because i think its better
went back to injecting because the morning routine was too fragile around shift work
went back to injecting
anyone switched from injectable to oral and kept the same effect
SNAC does the work
[edited]took 13 weeks before i saw anything on oral, roughly twice what the injection took me
ok so PIONEER was the original oral semaglutide programme and OASIS looked at the higher oral doses
lost 25kg over 1 months on oral, slower than my injectable stretch, i stuck with it anyway
tea also does not count, in case anyone was hoping
the oral disappointed me on the mornings i couldnt manage 30 minutes fasted, which was most of them, i think
i do 6am with water and eat at 7, thats been my routine for 13 months
one percent is the number people quote and the spread around it is the actual story, happy to be corrected
follow up is orforglipron a peptide at all
plain water only, about half a glass, then nothing for 30 minutes. i follow it exactly
about one percent
roughly one percent gets absorbed, which is why the tablet strength looks nothing like an injectable dose, thats just me
does SNAC do anything other than get it through the stomach
quick one the non peptide orals are the interesting part of this channel now, not oral semaglutide
genuine question splitting a dose made the absorption problem worse rather than better in my own log
[edited]SNAC raises the local pH and protects it long enough to cross, thats its whole job
plain water only
took mine with breakfast once by accident and that week was a write off
