#reconstitution 2025-08-03

Sunday47 messages11 participantstimes are UTC
Highlights from this day
  • freeze_thaw — i swirl for about thirty seconds and then leave it. never had one fail to dissolve 22:33
  • chlorhex — while im here i write the mg/ml on the vial with a sharpie the moment i reconstitute it 22:36
  • snac_and_water — ok so anyone worked out a recon volume that puts 2.4mg on a round unit mark 22:53
  • septum_sal — follow up is a slightly cloudy solution ever ok or is that always a bin it 23:02
  • sharps_bin_sid — update from 12 months ago: switched to a single concentration and have not miscalculated since i was wrong about this last year: it is fine to draw cold, it just… 23:24
OS

a slightly collapsed cake is cosmetic. a puddle in the bottom is not

30mg in 3ml is 10mg/ml, so 5mg is 50 units. half the barrel, feels like a lot but its right

thats fine

does temperature of the bac water matter at all

not sterile water

coming back after 4 months, has the pinned recon volume advice changed

PP

if the maths gives you 13.7 units, change the volume. do not try to hit 13.7

FT

i swirl for about thirty seconds and then leave it. never had one fail to dissolve

nope, redo it

CH

while im here i write the mg/ml on the vial with a sharpie the moment i reconstitute it

😂8❤️1🧊6

my rule is one concentration for everything so i never have to think about it at 7am

one unit is 0.01ml. 100 units is 1ml. everything else in this channel follows from that

PP

gross powder mass is not net peptide. water and counterion are in that number

i use bac water even for a single-use vial because it stings less, which is not the reason it exists, thats just me

if you cannot read the mark confidently, your volume is too small. redo it at a lower concentration

the vacuum will pull it in. let it. fighting it is how you get a face full of aerosol

how long after reconstituting before its actually dissolved

SA

i keep a spare vial of bac water in the fridge purely so i never rush the maths, thats just me

👍4

ok so anyone worked out a recon volume that puts 2.4mg on a round unit mark

PP

bacteriostatic water for anything multi-use. sterile water has no preservative in it at all

LO

lyophilised powder is far more robust than people think. it is the solution that is fragile

VB

Testing queue: 2 submissions open, 50 awaiting dispatch.

CH

coming back to this how do you all measure a half unit, or do you just not

SS

follow up is a slightly cloudy solution ever ok or is that always a bin it

📉6😂18🧪8

pick the volume so your dose is a whole number of units. thats the entire trick

right so i was wrong about this last year: it is fine to draw cold, it just stings more going in

VB

Archive lookup: septum_sal first appears in this channel on 2025-04-12.

SS

do not add more bac water to an already reconstituted vial, you have lost track of the concentration

while im here dead space in a standard insulin syringe hub is small but it is not nothing over a month

the meniscus thing is real. read it at eye level or you are guessing by a unit either way

SB

label the date too. you will not remember which vial is which by week 21

PP

aim the diluent at the glass wall, not at the powder. it goes in gentler

one unit is 0.01ml

FT

5mg in 2ml gives 2.5mg/ml, 0.25mg is 10 units. good for a starting dose

clear or bin it
