#reconstitution 2026-01-24

Saturday67 messages13 participantstimes are UTC
Highlights from this day
  • stable_not_volatile — 30mg in 3ml is 10mg/ml, so 5mg is 50 units. half the barrel, feels like a lot but its right 12:58
  • lyophile_liv — a 3ml syringe for the diluent and a u100 for the dose is the setup most people here end up with 14:23
  • sharps_bin_sid — the vacuum will pull it in. let it. fighting it is how you get a face full of aerosol 15:07
  • first_and_last_four — do i swirl for a specific length of time or just until its clear 17:52
AP

is the meniscus reading actually a real source of error or am i overthinking

[edited]
SN

right so my vial pulled the water in on its own, is that the vacuum or did i do something
the last few units are a real loss. i accept it rather than tilting the vial and drawing air

bac water or sterile water for something im going into weekly for 3 weeks

AP

if the maths gives you 13.7 units, change the volume. do not try to hit 13.7

thats 5mg/ml

AP

label the date too. you will not remember which vial is which by week 20

AP

a slightly collapsed cake is cosmetic. a puddle in the bottom is not, we shall see

AA

i write the mg/ml on the vial with a sharpie the moment i reconstitute it
benzyl alcohol is what makes it bacteriostatic. it is about 0.9% typically

SN

update from 26 months ago: switched to a single concentration and have not miscalculated since

SN

30mg in 3ml is 10mg/ml, so 5mg is 50 units. half the barrel, feels like a lot but its right

bacteriostatic water for anything multi-use. sterile water has no preservative in it at all, i could be wrong

quick one it foamed. is it ruined

my 10mg vial weighed 11.4g on the jewellery scale, is that wrong

HH

coming back to this do not add more bac water to an already reconstituted vial, you have lost track of the concentration

HH

i keep a spare vial of bac water in the fridge purely so i never rush the maths, ask me again in a month

TO

20mg in 2ml is 10mg/ml. 1mg is 10 units, 2mg is 20 units, easy

my rule is one concentration for everything so i never have to think about it at 7am

SN

lyophilised powder is far more robust than people think. it is the solution that is fragile
label the date too. you will not remember which vial is which by week 31

is a slightly cloudy solution ever ok or is that always a bin it

weighing the vial before you reconstitute is the only way to catch an underfill

dead space in a standard insulin syringe hub is small but it is not nothing over a month

i use bac water even for a single-use vial because it stings less, which is not the reason it exists

😂6📉3
AP

is it normal for the cake to look slightly collapsed on one side

TO

if you cannot read the mark confidently, your volume is too small. redo it at a lower concentration

LL

a 3ml syringe for the diluent and a u100 for the dose is the setup most people here end up with

🙏1😂15

nope, redo it

🧊1🤝8👍2
VV

im getting bubbles in the barrel every single time, what am i doing wrong

can i add more bac water later to a vial ive already reconstituted

does it matter if i inject the bac water straight at the powder

ok so 5mg in 2ml gives 2.5mg/ml, 0.25mg is 10 units. good for a starting dose

is it worth buying a 3ml syringe for the diluent or is the insulin one fine

too dilute imo

🤝61😂1

sorry to jump in swirl it. do not shake it. the foam is denatured protein and you are not getting it back

SB

the vacuum will pull it in. let it. fighting it is how you get a face full of aerosol

SB

whats the smallest volume you would trust yourself to measure on a u100

⚠️1❤️5🤝1
FT

follow up the jewellery scale is a good habit but calibrate it, they drift badly

SB

right so it should be completely clear with no floaters. cloudy means stop

KF

anyone worked out a recon volume that puts 10mg on a round unit mark

if you draw up with a bigger pin and inject with a fresh one you lose a bit more to dead space

why does everyone say 2ml specifically and not 1ml

do i swirl for a specific length of time or just until its clear

🎉1💀4🔥12
TF

nobody here should be telling you a dose. we can check your arithmetic, thats a different thing, thats one data point

TF

if the powder is on the stopper, tap it down before you add anything. it will still dissolve either way

do you let it come to room temp before drawing or draw it cold

TO

i was wrong about this last year: it is fine to draw cold, it just stings more going in, n of 1 obviously
