Highlights from this day
  • void_volume — follow up i stayed on 1.7 for 24 months and never went to 2.4, appetite was already where i needed it, thats one data point 19:11
  • orfor_watch — the plateau hit me at 34 weeks and moved again about a month later without a dose change 19:22
  • monoisotopic — went to 2.4 and the extra suppression wasnt worth the GI for me, so i came back down 20:10
  • two_ml_two — FLOW was the kidney outcomes trial and it gets quoted in here for things it didnt measure 20:30
  • two_ml_two — my PeptideMeter report on the FGP vial came back 98.1 against a certificate saying 99.4 21:12

did anyone titrate slower than four weeks a step and how did that go

going slow cost me nothing except time and saved me two bad weeks, thats my whole contribution

MO

SELECT was cardiovascular outcomes in people with overweight and existing disease, not a weight trial, ymmv

VV

lost about 3kg on the low doses before i even got to 1, which surprised me

VB

Digest for the week of 2024-09-16 has been published.

VV

genuine question does the licensed ceiling for weight differ from the diabetes one

OW

unrelated but the long half life flattens the trough more than people expect, i never felt a day seven dip, thats one data point

my plateau was the scale stopping while appetite stayed suppressed, which is a different problem

the plateau hit me at 34 weeks and moved again about a month later without a dose change

Once-Weekly Semaglutide in Adults with Overweight or Obesity and Type 2 Diabetes (STEP 2)
The Lancet · 2021
The type 2 diabetes population, where the weight response is generally smaller.
MO

restarted after 7 months off at a low dose and the nausea arrived exactly like the first time

🎉1⚠️1

my 2 vial in 1ml gives 8mg/ml so 5 lands on 12 units, thats why i picked that volume
moving my dose day by a day or two never did anything noticeable for me

AA

how long did the first 42kg take for people at the low doses

AA

compounded and brand felt the same to me at 1.7, which is one person and nothing more

[edited]

is there a reason to hold at 1.7 rather than going to 2.4

AA

five week steps worked better for me than four, purely on how the second week felt

MO

i was wrong about the ceiling for a year, i thought 3 existed. it doesnt for the weight indication

MO

went to 2.4 and the extra suppression wasnt worth the GI for me, so i came back down

changed my mind on the four week rule, i think it depends entirely on how the current dose feels
the trials titrated on a fixed schedule, most people here dont, worth remembering when comparing

how long till steady state, ive read the half life is about a week
changed my mind on the four week rule, i think it depends entirely on how the current dose feels

WZ

right so anyone gone from 12 straight to 15 or is that too big a jump

TM

FLOW was the kidney outcomes trial and it gets quoted in here for things it didnt measure

wk 1-4 0.25mg
wk 5-10 0.50mg (held 2 extra weeks)
wk 11-14 1.00mg
wk 15-26 1.70mg (held, long)
wk 27+ 2.40mg
SS

STEP 4 had the withdrawal arm, thats the one people quote when they talk about stopping

vials not pens here

[edited]

did anyone hold at 1 longer than four weeks before going up

TM

my PeptideMeter report on the FGP vial came back 98.1 against a certificate saying 99.4

SS

STEP is several trials, so saying STEP said x is usually wrong without a number after it

⚠️5

$240 a month for compounded against what brand costs locally is why most people here use vials

plateaus move

steady state takes weeks

licensed ceiling for weight is 2.4 weekly, anything above that is not a licensed dose

FI

for the archive im 10 months in at 12 and the appetite effect faded, is that a thing

TM

is the plateau at 11 weeks normal or am i doing something wrong

is 2.4 the top or do people go past it

AA

held at 0.5 and lost 89lb over 22 months, slower than the trials and fine by me
