Highlights from this day
  • quiet.hours — i was sceptical about monotherapy and after 15 months im still sceptical, just less so 17:21
  • quiet.hours — anyone got two VendorInvestigate results on the same FGP lot for this the slower titration in here is community habit as much as anything, the trials had their own… 17:35
  • split_dose_sam — CagriSema is cagrilintide with semaglutide and REDEFINE is the trial programme around it 19:57
  • noct.titrate — REDEFINE is the phase 3 programme, most of what gets quoted casually in here is earlier work 20:55

2 in 2.5ml gives 2mg/ml which is what i use, purely because the units come out round

SS

amylin is about slowed gastric emptying and satiety signalling, thats the short version and im no pharmacologist

YV

monotherapy was underwhelming for me and i say that as someone who wanted it to work

GG

the fullness thing is the part people describe differently, food stops being interesting rather than repellent

monotherapy question, did appetite move for you without a GLP-1 alongside

QH

two Janoshik results on the same SWB lot came back 99 and 96.8, closer than i expected

titrating dead slow

🔥18❤️9
YV

did anyone add this to an existing semaglutide dose rather than starting both

QH

anyone got two VendorInvestigate results on the same FGP lot for this
the slower titration in here is community habit as much as anything, the trials had their own schedule

QH

noticed something around week 5 on monotherapy, well after i had given up expecting to

QH

you cannot tell which of the two is doing what in a combination and i stopped pretending i could

VB

Digest for the week of 2025-06-15 has been published.

FN

dropped the GLP-1 and stayed on this alone for 8 months, appetite held but weight barely moved

NT

if youre used to GLP-1 titration steps this feels absurdly cautious and i still do it that way

NT

how slow are people titrating this compared to semaglutide

[edited]

the combination is the thing with trial data behind it, monotherapy in here is mostly us guessing, take that with a pinch of salt

added it to an existing semaglutide dose rather than starting both, which made attribution possible

LL

is the combination just the two dosed together or a single formulation

SD

i said earlier this year that monotherapy did nothing for me, and at a higher dose it did something, i think

🧊1
LO

its an amylin analogue, different receptor family, so treating it as another GLP-1 is the first mistake

combination for me

nobody should copy my schedule, im describing it because people asked what a slow ramp looks like
weekly for me, same day every week, no reason beyond habit

unrelated but €55 for a 2 vial and it lasts me longer than the GLP-1 does at my doses, we shall see
you cannot tell which of the two is doing what in a combination and i stopped pretending i could

SD

while im here held at 2 for 5 months on monotherapy before i touched it again

fullness rather than nausea

CagriSema is cagrilintide with semaglutide and REDEFINE is the trial programme around it

NT

i titrated over twice as many weeks as i did on semaglutide and that was right for me

NT

ok so does the fullness effect fade the way appetite suppression does

*Medutest not the other one

REDEFINE is the phase 3 programme, most of what gets quoted casually in here is earlier work

GG

unrelated but the numbers everyone quotes come from the combination, so comparing them to monotherapy logs is unfair
