Highlights from this day
  • tenpence — the numbers everyone quotes come from the combination, so comparing them to monotherapy logs is unfair 23:12
  • bo_bloodwork — i titrated over twice as many weeks as i did on semaglutide and that was right for me changed my mind on this, i started thinking amylin was the interesting bit and… 23:40
  • ghent_gradient — two VendorInvestigate results on the same Homopeptide lot came back 99 and 97.4, closer than i expected 23:58
GG

coming back to this nobody should copy my schedule, im describing it because people asked what a slow ramp looks like

right so the fullness thing is the part people describe differently, food stops being interesting rather than repellent

the combination is the thing with trial data behind it, monotherapy in here is mostly us guessing

different not less

TM

follow up 15 in 0.5ml gives 4mg/ml which is what i use, purely because the units come out round

TE

added it to an existing semaglutide dose rather than starting both, which made attribution possible

TM

coming back to this stopped for 1 months and the fullness went within a fortnight of the last dose

the nausea felt different rather than less, more fullness and less of the queasy wave

TE

GG

amylin is about slowed gastric emptying and satiety signalling, thats the short version and im no pharmacologist

underwhelming honestly

REDEFINE is the phase 3 programme, most of what gets quoted casually in here is earlier work

whats the recon people use, 10 in 0.5ml

BB

if youre used to GLP-1 titration steps this feels absurdly cautious and i still do it that way

TW

changed my mind on this, i started thinking amylin was the interesting bit and now i think its the pairing

TE

£185 for a 2 vial and it lasts me longer than the GLP-1 does at my doses

TW

i was sceptical about monotherapy and after 20 months im still sceptical, just less so, for what its worth

anyone holding at 1.7 for a while before stepping

how do people separate which of the two is doing what in a combination

i titrated over twice as many weeks as i did on semaglutide and that was right for me

GG

you cannot tell which of the two is doing what in a combination and i stopped pretending i could
