why does everyone say 2ml specifically and not 1ml
#reconstitution 2025-10-05
- tarpit_tam — both are defensible. what is not defensible is switching between them mid-vial and forgetting which one you are on 16:19
- swirl_not_shake — which brings me to the real question. this one is a Hubei Jianxiang vial and the fill looks light to me 17:14
- tarpit_tam — Janoshik for content and purity, yes. send a sealed vial, not one you already punctured 17:41
- swirl_not_shake — last one i had done, for reference. came back on mass 18:43
benzyl alcohol is what makes it bacteriostatic. it is about 0.9% typically, i could be wrong
15mg in 3ml is 5mg/ml. same numbers as 10 in 2, just more of it
ok so 10mg in 1ml is 10mg/ml, so 2.5mg is 25 units. thats the one i use
the vacuum will pull it in. let it. fighting it is how you get a face full of aerosol
a slightly collapsed cake is cosmetic. a puddle in the bottom is not
do i swirl for a specific length of time or just until its clear
how long after reconstituting before its actually dissolved
aim the diluent at the glass wall, not at the powder. it goes in gentler
read it at eye level
30mg tirz vial. what is everyone putting in these
3ml. 10mg/ml. every dose in the ladder is a whole number of units
clean the whole way up
30 mg + 3.0 ml = 10 mg/ml
2.5 mg -> 0.25 ml -> 25 u
5.0 mg -> 0.50 ml -> 50 u
7.5 mg -> 0.75 ml -> 75 u
10.0 mg -> 1.00 ml -> 100 u <- a whole barrel, split it or use two pinsthe 10mg dose being an entire barrel is the annoying part
i run 30mg into 1.5ml, so 20mg/ml, and take 12.5u for 2.5mg. half marks but a much smaller sting
both are defensible. what is not defensible is switching between them mid-vial and forgetting which one you are on
how do you keep track
write the concentration on the vial with a paint pen the second you mix it
mg/ml and the date. two things, ten seconds
ok
the powder itself takes up volume as well. 30mg of tirz is a visible cake
a few tens of microlitres at that mass. that is below your measurement error, ignore it
if you are working with 100mg and up it starts to matter
which brings me to the real question. this one is a Hubei Jianxiang vial and the fill looks light to me
light how
the cake is visibly thinner than the last two i had
cake height tells you almost nothing. change the lyophilisation cycle and you change the density
if you want to know the mass, weigh it or get it assayed. eyeballs do not do mass
Janoshik then
Janoshik for content and purity, yes. send a sealed vial, not one you already punctured
Medutest has been quicker for me lately on the identity side
different services, both fine. do not average their numbers, read them separately
and PeptideMeter is where i look when i want a vendor history rather than one vial
does the vendor matter for recon at all
for recon, no. for what is in the vial, obviously
GL Biochem powder i had years ago dissolved slower than anything since and i never worked out why
residual moisture and particle size. slow to dissolve is not the same as bad
last one i had done, for reference. came back on mass
and if this fill genuinely is light, the assay will say so in mg rather than in vibes
sending it monday
learning a lot in here
