#reconstitution 2026-03-18

Wednesday41 messages11 participantstimes are UTC
Highlights from this day
  • deadspace — follow up 30mg in 3ml is 10mg/ml, so 5mg is 50 units. half the barrel, feels like a lot but its right 21:00
  • deadspace — pick the volume so your dose is a whole number of units. thats the entire trick, ask me again in a month 21:05
  • meniscus_mel — a 3ml syringe for the diluent and a u100 for the dose is the setup most people here end up with 21:14
  • meniscus_mel — is it worth buying a 3ml syringe for the diluent or is the insulin one fine 21:53
  • meniscus_mel — how long after reconstituting before its actually dissolved 22:12
MW

aim the diluent at the glass wall, not at the powder. it goes in gentler

the meniscus thing is real. read it at eye level or you are guessing by a unit either way

MM

i keep a spare vial of bac water in the fridge purely so i never rush the maths

MM

is a slightly cloudy solution ever ok or is that always a bin it

thats fine

why does everyone say 2ml specifically and not 1ml

round numbers

DE

does it matter if i inject the bac water straight at the powder

🧪1

clear or bin it

NN

bac water or sterile water for something im going into weekly for 2 weeks

DE

follow up 30mg in 3ml is 10mg/ml, so 5mg is 50 units. half the barrel, feels like a lot but its right

too dilute imo

MM

a 3ml syringe for the diluent and a u100 for the dose is the setup most people here end up with

🧪3⚠️18

anyone else find the 30mg vials harder to get fully dissolved than the small ones

SN

my vial pulled the water in on its own, is that the vacuum or did i do something

if you cannot read the mark confidently, your volume is too small. redo it at a lower concentration

how much do i lose to dead space with a 30G pin

MM

lyophilised powder is far more robust than people think. it is the solution that is fragile, for what its worth

VB

Inter-lab diff for lot B-0114: 99% vs 97.4%. Within expected range.

VB

Digest for the week of 2026-02-15 has been published.

MM

is it worth buying a 3ml syringe for the diluent or is the insulin one fine

10mg vial
  + 1.0ml  -> 10.0 mg/ml   0.5mg =  5u   1mg = 10u
  + 2.0ml  ->  5.0 mg/ml   0.5mg = 10u   1mg = 20u
  + 2.5ml  ->  4.0 mg/ml   0.5mg = 12.5u (dont)
  + 5.0ml  ->  2.0 mg/ml   0.5mg = 25u   1mg = 50u
VB

Purity check: no report on file for lot G-0641. Nothing logged either way.

MM

how long after reconstituting before its actually dissolved

MW

do not add more bac water to an already reconstituted vial, you have lost track of the concentration

TT

is the meniscus reading actually a real source of error or am i overthinking

RT

gross powder mass is not net peptide. water and counterion are in that number
if the maths gives you 13.7 units, change the volume. do not try to hit 13.7

RT

update from 20 months ago: switched to a single concentration and have not miscalculated since

it should be completely clear with no floaters. cloudy means stop, still working it out
i was wrong about this last year: it is fine to draw cold, it just stings more going in
