steady state is the bit people skip, judging a new dose after eight days tells you almost nothing

- assay_not_purity — sorry to jump in the plateau hit me at 23 weeks and moved again about a month later without a dose change 12:29
slower worked better
what did SELECT actually measure, i see it quoted for everything
going slow cost me nothing except time and saved me two bad weeks, thats my whole contribution, happy to be corrected
im at 0.5 and stuck, did anyone break a plateau without going up
is 2.4 the top or do people go past it
SELECT wasnt weight
plateaus move
STEP 1 ran to 68 weeks. thats the one everyone half remembers
thats STEP 1
update on the earlier thing changed my mind on the four week rule, i think it depends entirely on how the current dose feels
genuine question i held at 0.25 for 8 months and it was the best decision i made
if youre comparing brand to compounded, dose accuracy in the vial is the variable, not the molecule
STEP 4 had the withdrawal arm, thats the one people quote when they talk about stopping
went 1 to 1.7 too fast and spent two weeks regretting it, dropped back and repeated the step
mine faded too
sorry to jump in did anybody find a difference between QST and pharmacy at the same 7.5
right so dose day is sunday for me only because thats when i remember, not because sunday matters
follow up my VendorInvestigate report on the ERP vial came back 96.8 against a certificate saying 98.6
wk 1-4 0.25mg
wk 5-10 0.50mg (held 2 extra weeks)
wk 11-14 1.00mg
wk 15-26 1.70mg (held, long)
wk 27+ 2.40mgwent too fast once
my 15 vial in 2ml gives 2mg/ml so 0.5 lands on 8 units, thats why i picked that volume
i dont think the plateau is a dose problem most of the time, but i cant prove that
went to 2.4 and the extra suppression wasnt worth the GI for me, so i came back down
seven days ish, thats just me
Reminder set. I will post here in 6 days.
2.4 is the ceiling
sorry to jump in the plateau hit me at 23 weeks and moved again about a month later without a dose change
my dose day drifted from friday to sunday, does that reset anything
FLOW was the kidney outcomes trial and it gets quoted in here for things it didnt measure, anyway
wk 1-4 0.25mg
wk 5-10 0.50mg (held 2 extra weeks)
wk 11-14 1.00mg
wk 15-26 1.70mg (held, long)
wk 27+ 2.40mghow many weeks before you noticed anything at 5
whats the actual licensed ceiling, i keep seeing different numbers
give it four weeks
half life is about a week so youre roughly four to five weeks to steady state on any new dose
is there a reason to hold at 1.7 rather than going to 2.4
moving my dose day by a day or two never did anything noticeable for me
does the seven day half life mean the last two days are weaker
the appetite effect faded for me around 3 months and going up fixed it for a while
same dose day
| Trial schedule | What most people here do | |
|---|---|---|
| Step interval | 4 weeks, fixed | 4-12 weeks, on symptoms |
| Holding | protocol deviation | normal and expected |
| Top dose | reached by design | often never reached |
| Coming down | not studied | common at maintenance |
was SURMOUNT-OSA the one that ran to 68 weeks or am i mixing them up
while im here i was wrong about the ceiling for a year, i thought 3 existed. it doesnt for the weight indication, happy to be corrected
SELECT was cardiovascular outcomes in people with overweight and existing disease, not a weight trial
not the same trial
vial units and pen clicks are different systems, mixing the two is how people end up wrong
5 months at 2.4 and the honest summary is diminishing returns after the first half, thats just me
held at 10 and lost 75lb over 11 months, slower than the trials and fine by me, still working it out
steady state takes weeks
how long till steady state, ive read the half life is about a week
i stayed on 1.7 for 21 months and never went to 2.4, appetite was already where i needed it
£120 a month for compounded against what brand costs locally is why most people here use vials
compounded and brand felt the same to me at 10, which is one person and nothing more
im 18 months in at 1 and the appetite effect faded, is that a thing
restarted after 25 months off at a low dose and the nausea arrived exactly like the first time
lost about 41kg on the low doses before i even got to 1, which surprised me
vials not pens here
five week steps worked better for me than four, purely on how the second week felt
is the plateau at 22 weeks normal or am i doing something wrong
does moving my dose day by two days matter with a seven day half life
does the licensed ceiling for weight differ from the diabetes one
does anyone actually run a five week step instead of four
my plateau was the scale stopping while appetite stayed suppressed, which is a different problem
right so licensed ceiling for weight is 2.4 weekly, anything above that is not a licensed dose
how did people handle the jump from 1 to 1.7
held there too
went too fast once, i think
went too fast once
how long did the first 47kg take for people at the low doses
